Description Deep functional profiling of immuno-oncology markers and cytokines in whole blood (WB) reveals immune signatures that inform immunotherapy selection and patient outcomes in cancer research. WB analysis is time sensitive, and collection and cytometric analysis often occur at different sites, leading to significant delays. This challenge can be addressed using WB stabilizers. We developed modular Flex-Fit CyTOF™ panels compatible with WB stabilizers to enable flexible workflows and rapid panel design. High-parameter flow cytometry mediated by CyTOF technology enables single-tube staining to identify 50-plus markers at the single-cell level. CyTOF technology provides exceptional resolution without spectral unmixing or compensation. Two high-parameter antibody panels – the Checkpoint CyTOF Panel (21 markers; identifies 12 immuno-oncology markers on 10 major cell subsets) and the Cytokine CyTOF Panel (31 markers; identifies 10 cytokines on 32 cell subsets) – were developed for analysis of fresh and stabilized WB. Following WB stabilization, the analyzed cell populations maintained the anticipated co-expression of markers and had no nonspecific staining. Taken together, Flex-Fit CyTOF panels and flexible CyTOF workflows enable fast and convenient high-parameter cytometric analysis of WB in cancer research. For Research Use Only.
